DAS, INDRAJIT. Effects of heart work and insulin on gbcogen metabolism in the perfused rat heart. Am. J. Physiol. 224(l): 7-12. 1973.-A study was made in the isolated perfused rat heart of key enzymes for the synthesis and breakdown of glycogen and the major intermediates on the glycogen pathway. Hearts from normal animals, animals rendered insulin deficient by injection of antiinsulin serum, and animals made diabetic with streptozotocin were perfused both retrogradely via the aorta (Langendorff) and atrially, while they were pumping liquid against a hydrostatic pressure head (working), with and without insulin in the perfusate. Hexokinase isoenzyme type II was reduced by 35% in streptozotocin diabetic hearts. Heart work also significantly reduced the normal hexokinase isoenzyme type II. Insulin prevented the loss of hexokinase isoenzyme II in the *working heart, but it did not have any effect on the diabetic perfused hearts. Total glycogen synthetase activity was affected neither by heart work nor by insulin deficiency. Heart work increased the glycogen synthetase I/D (active/inactive) ratio in both normal and diabetic hearts. Insulin in the perfusate was unable to increase the I/D ratio in working hearts, but insulin in vivo increased the I/D ratio significantly within 30 min in diabetic hearts. Phosphorylase activity was not affected by heart work or insulin. Significant increases of the tissue contents of glucose 6-phosphate, glucose 1 -phosphate, and UDPglucose and glycogen were observed in both Langendorff and working hearts of normal rats with insulin in the perfusate.

Role of Fructose as a Potent Antiarrhythmic and Anti-infarct agent in Isolated Rat Heart

In the current study, effects of acute short term administration of fructose on cardiac arrhythmias and myocardial infarction size following ischemia/reperfusion were investigated in isolated rat heart. The hearts were subjected to 30 min zero flow global ischemia followed by 120 min reperfusion. In the control group, the hearts were perfused by normal drug free Krebs-Henseleit (K/H) solution t...

Effects of L-Carnitine on Cardiac Apoptosis in Ischemic- Reperfused Isolated Rat Heart

Carnitine is a vital biologic substance for transporting fatty acids into myocytes. It also facilitates fatty acids β-oxidation for energy production. In this study, effects of L-carnitine (L-Car) on apoptosis in the ischemic isolated rat heart were investigated. Male Sprague-Dawley rats were divided into four groups and anesthetized by sodium pentobarbital. The heart was removed and mount...

Effects of Teucrium polium Ethylacetate Extract on Serum, Liver and Muscle Triglyceride Content of Sucrose-induced Insulin Resistance in Rat

Possessing putative hypolipidemic effects, Teucrium polium (TP) have been traditionally used as a medicinal plant in Iran. The aim of the present study was to investigate this effect on the sucrose-induced insulin resistance male rat model. Thirty Wistar male rats weighting 180 ± 20 g were divided into five groups of six each. Four groups were given sucrose 50% in drinking water for 10 weeks. I...

EFFECT OF INSULIN IN THE PRESENCE OF GLUCOSE AND AFTER DEPLETION OF GLUCOSE, GLUCOSE 6-PHOSPHATE AND GLYCOGEN By ERNST B. CHAIN and PETER M. SENDER*

In the absence of glucose, insulin stimulated the incorporation of 14C-labelled amino acids into protein by perfused rat hearts that had been previously substantially depleted of endogenous glucose, glucose 6-phosphate and glycogen by substrate-free perfusion. This stimulation was also demonstrated in hearts perfused with buffer containing 2-deoxy-D-glucose, an inhibitor of glucose utilization....
